49- Recombinant DNA Technologies

Covers the core techniques of recombinant DNA technology, starting with the combining of DNA from different sources using vectors and host cells. It then moves through restriction endonucleases (Type I and Type II), reverse transcriptase and cDNA synthesis, and methods for identifying sequences such as gel electrophoresis and Southern blotting. Cloning and PCR are described next, followed by Sanger dideoxy sequencing and next generation sequencing platforms including Illumina, pyrosequencing and Ion Torrent, with their applications.

Recombinant DNA Technologies

Recombinant DNA (rDNA) technology involves manipulating DNA to create new genetic combinations. These techniques are foundational for modern biotechnology, enabling advancements in medicine, agriculture, and biological research.

Recombination of DNA

Definition : Combining DNA from different sources to form new genetic constructs.

Process :

  • Isolate DNA fragments.
  • Use vectors (e.g., plasmids) to insert DNA fragments into host cells.
  • Host cells replicate, producing multiple copies of recombinant DNA.

Role of Key Enzymes and Methods

Restriction Endonucleases:

  • Function : Enzymes that cut DNA at specific sequences, called recognition sites.

Types :

  • Type I: Cuts DNA far from the recognition site.
  • Type II: Cuts DNA at specific locations within or near the recognition site (most commonly used).
  • Application : Generate DNA fragments for cloning, mapping genomes, and genetic engineering.

Role of Key Enzymes and Methods

Reverse Transcriptase:

  • Function : Synthesizes complementary DNA (cDNA) from an RNA template.
  • Applications :
  • Convert mRNA into cDNA for cloning and sequencing.
  • Study gene expression and produce recombinant proteins in eukaryotic cells.

Identifying DNA Sequences

Electrophoresis

  • Principle : Separation of DNA fragments based on size and charge.

Process :

  • DNA samples loaded into a gel matrix.
  • Electric current applied; DNA fragments migrate through the gel.
  • Smaller fragments move faster, resulting in separation by size.
  • Applications : Analyzing DNA fragments, checking PCR products, and preparing samples for further experiments.
